杨兴斌男,博士,汉族,现为教育部直属、国家首批“双一流”建设高校bat365在线平台官网院长、二级教授、博士生导师省部级劳动模范陕西省食品科学技术学会理事长1997年获北京化工大学精细化工工学硕士学位,2004年获空军军医大学(原第四军医大学)医学/药理学博士学位,澳大利亚西澳大学(University of Western Australia访问教授(食品营养与健康的临床研究)。入选陕西省中青年科技创新领军人才陕西省创新人才推进计划(科技创新团队)荣获全国模范军队转业干部称号近年持续入选全球前2%顶尖科学家榜单。

现任陕西省食品绿色加工与安全控制工程实验室主任陕西省农产品贮藏加工危害因子风险评估实验室主任西部果品资源高值利用教育部工程研究中心技术委员会副主任西北濒危药材资源开发国家工程实验室和药用资源与天然药物化学教育部重点实验室学术带头人。目前主要从事食物药物的肠道菌群代谢与营养、脑认知健康,功能食品与杂粮食疗(药膳)食品创制,以及茯砖茶等发酵茶的加工及其金花菌等益生菌发酵功能(保健)食品和预包装食品的应用基础研究和产业化工作。

2. 主要学术贡献和研究方向

长期从事食物及药食同源植物的功能化学成分制备、化学结构表征和营养健康作用研究。针对目前慢性疾病高发和药物防治的副作用和安全性问题,筛选新型食源性和药食同源的安全和高效食品功能因子,阐释其代谢动力学和分子生物学的营养调节机制,开发多元化高附加值的功能性食品和健康产品。目前,已在食品营养与健康科学基础理论研究领域中取得以下代表性成果:

1创立了血管内皮舒张因子一氧化氮(NO)代谢物硝酸盐(NO3-)、亚硝酸盐(NO2-)与“NO”RSNOs的高灵敏度GC-MS和化学发光分析技术,为准确研究NO代谢及食品血管营养提供了国际领先水平的技术参考;发现生物样品预处理中清除干扰物NO2-的酸性磺胺可导致“NO”RSNOs分解而低估RSNOs测定值,并确立了最适磺胺浓度,澄清了长期以来备受争议的该分析方法低估NORSNOs的事实,为血管分子营养评价提供可靠指针,且发现长期被推测为致癌的限量摄入物硝酸盐(NO3-)、亚硝酸盐(NO2-)及富含NO3-的菠菜维持血管内皮功能稳态中的重要营养价值;针对当前F2-isoprostanes20-HETE等细胞膜花生四烯酸氧化损伤代谢物标志分子的GC-MS 分析技术缺陷进行了方法学完善,并基于高灵敏度的UPLC-Q-TOF-MS/MSGC-MS分析手段,建立了脂肪酸代谢轮廓谱的研究方法;构建了同时检测氧化三甲胺(TMAO)、三甲胺(TMA)、二甲胺(DMA)的UPLC-MS/MS同位素标记分析方法,首次发现含三甲胺结构的卵磷脂等甲基供体营养素经肠道生理菌群代谢产生TMA及其进一步经“肠-肝轴”的肝脏含黄素单加氧酶3FMO3氧化为TMAO,进而诱导肝损伤危害,拓展了卵磷脂等营养素的营养与安全“利与弊”的科学认知。

2同时,系统研究L-阿拉伯糖、寡糖、多糖等功能糖与类黄酮的互作营养作用和分子调控机制,证实了类黄酮激活的小肠II相代谢酶(SULTUGT)和外排转运蛋白(P-gpMRP1)是其肠吸收抑制和低生物利用度的关键调控靶点,首次发现水苏糖等功能糖通过抑制小肠II相代谢酶(SULTUGT活性和外排转运蛋白(P-gpMRP1表达而拮抗类黄酮的肠道上皮细胞首过代谢(首过消除, 进入体循环之前的代谢),提高类黄酮(含异黄酮)的肠吸收和生物利用度,揭示功能糖与类黄酮的营养互作新功能和新机制,并发展了食品营养物质基础整体观的网络食品功能学理论和研究新技术;发现L-阿拉伯糖、水苏糖和大豆多糖等功能糖提高的肠道琥珀酸小分子代谢物能够激活结肠上皮细胞果糖-1,6-二磷酸酯酶(FBPase)和葡萄糖-6-磷酸酶(G6Pase)而促进结肠糖异生(Intestinal gluconeogenesisIGNIGN升高的葡萄糖在肠道门静脉被激活的葡萄糖感受器蛋白SGLT3感知,并经--轴反射性提高大脑神经元细胞活性蛋白C-FosCGRP的表达,反馈性抑制肝脏糖异生(Hepatic gluconeogenesisHGN,进而调控机体的血糖稳态,构-效关系研究揭示大分子多糖较L-阿拉伯糖、水苏糖呈现更强的效应

3此外,我们发现茯砖茶多糖较其原料黑毛茶多糖有显著的脂肪产热活性,并发现冠突散囊菌及其胞外多糖通过肠道菌群依赖性的吲哚乙酸(I3A)、吲哚丙酸(IPA)等色氨代谢物靶向脂肪细胞AhR-FGF21-UCP1芳香烃受体信号通路调控脂肪产热的分子营养新功能与新机制我们惊讶的发现,水苏糖与小肠上皮细胞膜上的HSP90β蛋白结合改变其外泌体miRNA谱,首次揭示了低聚糖在上消化道的新功能,修正了寡糖益生元是小肠过客的传统认知;阐明了水苏糖-小肠miRNA-结肠菌群的轴向调控关系,揭示了水苏糖下调的小肠miR-30a-5p显著抑制结肠/粪便Lactobacillus reuteri增殖的间接益生新机制,证实了水苏糖作用小肠上皮细胞膜HSP90β调节外泌体miRNA谱的新功能和新机制。
